Homemade HBSS test
The last couple of posts showed that homemade HBSS does not cause discoloration, even though commercial HBSS does. I wanted to test if there was something mysteriously special about this solution, so I looked at non-RTP’ed wafer 622 in the incubator in solutions with and without serum (10% FBS). Even without serum, the cell media caused rapid discoloration. Again, homemade HBSS discolored slowly, but there was complete discoloration in HBSS + 10% FBS after 5 days. Serum is too complicated to tear apart its components, but it seems to speed up discoloration. This is the first homemade HBSS-containing solution that caused discoloration, so at least there’s nothing magical about the chemicals in our lab.
