Membrane collapse in culture media
Above video shows the disruption of the membrane from W404 after around 12 hours in the DMEM media (benchtop). The membrane was not treated with UV-Ozone and degraded pretty early as compared to that from W336 which broke at ~28 hrs. The membranes were loaded with 15um beads to check if they were intact and…
Permeability and Convection/Diffusion
Permeability data first:Wafer 310r = 4.3 units from centern = 32.0 mg/mL cytochrome C in 1x PBSmean permeability = 27-33 uL/(min-cm2-psi) *** corrected to account for (2) 115x2014um slits[The range is due to variations in measurement and total accounting for initial uL]Wrapping with parafilm resulted in only a mean of 13uL of 1000 initial uL…
TEM Tomography of NPN and npMgF2
Many, many thanks to Kilean Lucas and Brian McIntyre who enabled our metrology. We have desired to get 3d information about the structure of our nanopores. One method to do so is known as electron tomography, a technique which can reconstruct volumes and surfaces from sets of tilted images. By identifying dense regions of contrast…
Theoretical Underpinnings of Small Molecule Permeability Measurements in the µSiM (Part 2: Experimental Validation)
ByJimThis is the second installment in a series of posts focused on the development of a method for measuring barrier permeability in the µSiM. In Part 1 we discussed the goal of making these measurements in situ at a position beneath the membrane. Because our preference is to use the trench down configuration in the…
Summary of COMSOL simulations for Endohm TEER measurements
This is the final post (hopefully) summarizing my simulation results I did to model Endohm system. Barrett used endohm cup system to do the coculture of bEnd3 and glial cells for his BBB studies. The resistance values obtained while using pncSi and uniformly porous membrane for cell growth were drastically different, and it was not very…
SEM images for Micro & Nano Scale experiments
ByAslanHello Everyone, I have been trying to take SEM images in 3 different steps during my experiments with polystyrene beads over Track Etched membranes including before capturing, After capturing and releasing. Micro Scale Experiments: As you might remember, in Micro-scale experiments, we use 10 micron beads and 8 micron pores. Before capturing: This is only…

Seems 187 was a bad choice for these experiments ( see Maryna’s post). Clearly transport through the same membrane can be possible by diffusion (Jess’ diffusion experiments with 187 worked) and impossible by convection.
This failure also illustrates the usefulness of posting data shortly after it comes in. I don’t know when Maryna had this data, but if it could have been posted before Thursday we might have picked another membrane.
Can these experiments only be done in small sets? It would be useful if 2-4 samples from each new wafer could be run to test flow rate.
If the EQ experiments are really cumbersome, we need to find a way to make them easier, or they will provide very little near-term feedback. What is the bottleneck?
