BSA-DDM Complex Intensity
ByMike HI took a look back through the Melvern data I collected, particularly focusing on changes in the intensity data in the BSA-DDM solution (the previously posted plots were of number data). When I compared the peak intensities for BSA, DDM, and DDM-BSA complex I noticed that the intensity peaks fall over the same size distribution…

HUVECs & Microscopy with transwells
Recently, I’ve been frustrated with polycarbonate transwells since they are opaque and I couldn’t see cells on them. So, I bought new transwells from BD (Falcon, PET membrane, 0.4um pores) and decided to test these out. I also wanted to see if coating the transwells with gelatin would promote cell adhesion. These are P6 HUVECs…
Cell Culture on Gradient Microporous-NPN™
ByAlecIntroduction As a continuation to my previous post, I am running a triplicate of cell culture adhesion tests to observe cell lose on our membranes following physiological shear stress. Methods HUVECs (P3,4,5) were seed within our mimetic and left to grow to confluent for 24 hours. Shear (4.5 dyn/cm2) was initiated and images were collected…
Getting linear standard fluorescent curves with the Tecan
Now that we have preliminary transport data from the Tecan, we need standard curves in order to get permeability coefficients. This is really tricky because the RFU (relative fluorescent units) are arbitrary and based on the Tecan gain. For example, a solution of 10uM rhodamine might be 5000 RFU with a gain of 80 but…
TECAN absorbance for 96-well plates is topping out even with pure buffer
EDIT: This problem was solved thanks to Tejas. It turns out that Falcon BD transparent 96 well plates absorb in the UV range, and are best used for bradford assays or anything that measures absorbance higher than the UV range. Apparently they sell UV-inactive 96 well plates, but they are very expensive. Josh Winans and…
