HUVEC spreading on pncSi and glass
Following time lapse movies show the spreading of HUVECs on pncSi and glass ( @ 37C, in L15 media supplemented with 10% FBS) over a time period of ~4 hrs.

A quick recap: Vaccinex has asked the lab to design a closed system device with the capability to co-culture on microporous membranes. This device would be used to mimic the blood brain barrier, with endothelial cells cultured on the apical side and glial cells cultured on the basal side, and would allow for the testing…
Last week when I reported no membrane breakage after a week of HUVEC culture, I showed a phase micrograph on day 7. This week I tracked the HUVECs a bit more closely with the microscope. This is 10X, HUVEC P5, initial seeding density of 32,500 cells/insert. I tried to capture approximately the same area on…
The goal of this project is to determine the viability of PEG as a structural support for large area nano porous membranes. This would enable lift-off vs. the current method of etching through the wafer. The ideal properties are that the structure can withstand an aqueous solution for many days without peeling off or dissolving,…
A little while back I posted a design for a capture device that would (nominally) work for dialysis on real patients. Due to strict requirements to avoid clotting, the design included a lot of curves and nuanced structures that would be no problem for a lithographical or molding method, but which are a step too…
I tried to measure the thickness of the non-carbonized and carbonized membranes for the paper using SEM. To have a clear cut of the membrane I place the membrane chip upside down on a silicon wafer piece, then bend the membrane chip with tweezers so the membrane sticks to the wafer while breaking of the…
I was having trouble using the spectrometer to measure rhodamine concentration in the stir cell. I ran a standardized absorbency of the serial dilution of rhodamine. The curve is posted below. It appears to be linear. The molarity of rhodamine must be above 2uM in order for the spectrometer to read it. I think I…

Looks great.
So we have fibroblasts on glass and HUVEC on pnc-Si. I guess we have a couple more to go?
Make sure you can submit your to the publisher at the same play rate and same duration to make the point that pncSi = glass when it comes to cell spreading.
Yes, I am left with HUVEC on glass and fibroblast on pncSi. I tried fibroblast on pncSi, but it doesn’t look great as the seeding density was high.
I got that point and will make sure the movies are ready for submission.
