Sepcon-transwell discoloration II

I repeated a previous experiment but this time I used samples (no RTP) from W685 in oven @ 37C in DMEM(-FBS).

Four type of samples (blank chip with no slits, chip with both intact membranes, chip with one intact membrane and chip with both broken membranes) were used at different time points.

Again the expected trend [discoloration order (faster to slower): both broken membranes > one intact membrane > both intact membranes > blank chip with no slits] wasn’t observed. But similar trend as the first study could be seen in the following figure. The sample with both intact slits discolors the fastest! Also, the ‘one slit intact’ sample had few cracks in the membrane whereas the membranes in ‘both slits intact’ sample were completely broken.

Is there some kind of interface effect that could take place between the free standing membrane and the supporting material that triggers this process?
