RTP + Silanization

After recent studies with surface treatments, we wanted to take a step further by trying amino silanization on RTP’ed samples. Wafer 413 samples were RTP’d with no intact slits. Both 2D and 3D formats were tested in DMEM (-FBS) in oven @37C. All 3D samples had membrane side facing down whereas, in 2D format the membrane side was up. I followed the same silanization protocol as last time.

[Due to unexpected circumstances, I had to leave all the samples (in media) on benchtop overnight (12 hours) after keeping them in Oven @37C for 7 hours, to slow the discoloration, after which they were imaged for the first time point and again moved into the Oven]

As seen above, RTP+silanization helps to a greater extent and the treated sample (+RTP,+APTES) is not yet discolored. Whereas, discoloration of ‘silanized only (-RTP,+APTES)’ sample is slower than the ‘untreated control (-RTP,-APTES)’ sample but faster than ‘+RTP,+APTES’ treated sample.

The 3D format samples are still not discolored in the same time points.
