Optimization of HUVEC Network Formation in Geltrex On and Off Membranes
Our motivation was to look into how ADSC influence HUVEC network formation. My first objective was to investigate the optimal cell count, the best volume of Geltrex and if it was better to use M200 media with or without growth supplements. I then looked into seeding HUVEC on a nonporous membrane. At the same time,…
Auger Electron Spectroscopy: A Basic Overview
ByKileanSince I am posting data that is acquired using Auger electron spectroscopy, I thought that it might be a good idea to write a post about the theory of the instrument, to better understand why this data is useful and relevant. The Auger effect is a phenomenon in which an atom that is excited by…
eLIFE submission: A silicon nanomembrane-based in vitro platform to visualize immune cell trafficking across the human blood-brain barrier
ByJimWe have been collaborating with Britta Engelhardt’s group at the University of Bern in Switzerland since Tejas and I met them at the CNS Barriers conference in Paris in 2015. They are interested in studying live T-cell transmigration across human BBB models as part of their research program in Multiple Sclerosis (MS). Prior to working…
Tom's Latest Gold Separation Figure
ByJimI asked Tom to add gold PES separations of NPs to his earlier figure and to evaluate the loss of NPs during the use of these filters compared to pnc-Si. Here is what he came up … All experiments (including pnc-Si) were done at 10 PSI with 200 ul of starting material above the membrane…
More on porosity vs. position
Last week I took a series of TEM images from all but one position on wafer 638. Below are the TEMs, histograms, and porosities vs. distance from center plots. “Xpos”, “xneg”, “ypos”, “yneg” indicate which axis the images were taken from. Positive x-axis Negative x-axis Positive y-axis Negative y-axis Wafer 638 porosity vs. position While…

The brightness of the image may even be significant. Was the beam noticeably dimmer with the thick PVP coating. IF so, the film is very thick.
Anyway, the pores are clearly covered, so the fact that water passes so easily must mean that the film either dissolves or is highly porous. It will be interesting to see if there is an effect on separations.
