Cell Adhesion Testing 6/3-6/4

Two more adhesion tests were run recently, one on Tuesday and one on Wednesday. For both tests cells were imaged after five hours.

Fibroblast trial 3 was the first test where the optimal cell density (assuming 100% of cells are detached by trypsin and 100% attach to the substrate) was measured using a hemacytometer. Of note in this trial was the large error bars; from now on I plan on adding more PBS washing steps to remove any floating cells that may be present. I am skeptical of plastic w/o serum producing greater attachment; this may be partially due to my methods of seeding the cells. I have been seeding the cells in the plastic w/o serum rings last. The suspended cells may be settling in the flask, but I do constantly gently shake the flask to try to make it uniform. From now on I am randomizing the order of which I seed the cells onto each substrate to see if this changes anything. Two more fibroblast adhesion tests were started today, with results to be posted soon.
