Etaluma™ Microscopy: A Fully Incubate Microscope for Live-Cell Analysis and PMN Migration Videos

Introduction

Etaluma is a company that builds high resolution microscopes in a compact format for integration into cell culture incubators. This shifts from the traditional stage top or incubator cage approach to live cell imaging to allow for steady time-lapse imaging over extended periods of time without issues of focus or worries of temperature or CO2 fluctuations. With the recent development of Aline devices for live cell imaging, we believe this microscope will allow us to take the platform to the next level, and allow for high-content imaging experiments in a highly controlled environment. Etaluma agreed, and sent us a microscope to test for ourselves.

Cell Proliferation

HUVEC grown on NPN, 30 min post seed – 24h
HUVEC proliferation of NPN 24h – 48h post seed

PMN Migration

 

HUVECs, PMNs added luminally, 1 nM fMLP added abluminally, 10 fpm

Live VE-Cadherin

HUVECs were grown to confluency and stained with an external FITC-conjugated VE-cadherin (BV9) Ab. PMNs were added luminally and 1 nM fMLP was added abluminally. Time-lapse shows alternating phase/FITC at a 6 fpm
