BMES 2019 – Cell matrix interactions and platforms for creating engineered ECMs
BMES 2019 saw a number of talks being presented on the topic of controlling extracellular matrix architecture and understanding how cells respond to physical cues from their in ECM. My focus was primarily on talks pertaining to the tumor microenvironment or other tissues which show anisotropy in ECM fiber alignment such as cardiac tissue. Recently,…
Beads experiments with NPN membranes
ByAslanHello Everyone, For the next figure in our paper, and to make the transition happens smoothly from Track Etched membranes to NPN membranes, we decided to run the same beads experiment on NPN membranes as well. So Kilean and i started working on these experiments, and the first experiment was to capture and release 100…
Noncytotoxic dye SiR-DNA can be used for advanced automated tracking of cells
What is SiR-DNA? It is a far-red DNA stain, known as SiR-Hoechst, and can be used for live-cell imaging. Most current DNA dyes are toxic or affect the cell’s motion or proliferation. https://www.nature.com/articles/ncomms9497 It does not affect the migration of human dermal fibroblast cells (hdFb). The motility parameters (speed and displacement) for a 24 hour…
E-beam Deposition of Ag, Cold ALD Runs, and Sepcon Assembly For Making a Nanofluidic Transistor
Last week I conducted a variety of experiments to try to make a nanofluidic transistor (a device described in detail in this previous journal club post). I did some light optimization of the E-beam deposition of Ag films over a 3nm Ti adhesion layer in the PVD lesker machine and found that 12nm of Ag gives…
Fluorescent HUVECs in PC transwells
I wanted to do some more fluroescent imaging in transwells to look into background fluroescence. This was mainly motivated by my inital study. These are HUVECs stained with 5uM CMFDA and 10uM Hoechst 33342, fixed in formaldehyde and imaged on the Zeiss at 10X. This was 6 hours after seeding. Before acquiring the images, I…
Au Nanoparticles deposited on tented posts
Continuing my work from last week, Tucker used the same tenting (NPN, vapor transfer) and incubation procedure (20 nm and 60 nm Au particles) on an oxide substrate with posts (200 nm high). The substrate was dessicated in the 70C oven overnight. No additional platinum or gold coatings were made. The sample was tilted slightly…

So vapor salinization slows degradation but does not prevent it and L15 is just fine.
1) We’ll need to find more complete surface chemistry to work in general media.
2) We still need to do the ‘reconstitution’ studies to prove that it is a combination of bicarbonate and salts that causes the trouble in DMEM and other media – lets do this next please.
Anant,
If you vapor silane treat the surfaces – but don’t oxidize them beforehand with peroxide – what do you think is happening?
