More vacuoles on free-standing than supported pnc-Si (and PET)
One of the other suggestions from Angela Glading at lab meeting a while ago was to quantify the vacuoles on different culture areas. So, I selected images from 3 different experiments (all at day14 of growth) and counted the number of cells which expressed vacuoles on different areas. I then used my mask code in MATLAB to calculate the free-standing vs. supported pnc-Si area in each image and calculated the number of cells expressing vacuoles per unit area. I did the same for commercial PET inserts. Error bars are standard deviations.
So, there are significantly more vacuoles on free-standing than supported pnc-Si and PET. Into the paper….
