Discoloration – Covered edges and well side
Last week, I repeated Anant’s experiment which aimed to eliminate edge effects by painting over chips with nailpolish. I painted the entire well side of the chip but tried to keep the wells clear of polish (“painted back”) and I covered just the edge of a chip but tried to keep polish off the well wide (“painted edges”). A flat chip and a chip in the normal Sepcon format were controls. I used a different wafer (501) and different media (RPMI + 10% FBS) than Anant did.
Painting the well side and edges with nailpolish did not slow down discoloration – it may have actually sped it up. The flat chip has a weird pattern so I’m not sure if this actually discolored slower or not. The Sepcon format caused slightly slower discoloration (it looks like). I used fat O-rings in these experiments so the amount of chip exposed to media in the Sepcon format was smaller than usual.
