BSA Adsorption Studies 2
In our previous adsorption study, all of the surfaces seemed to adsorb a similar amount of protein. We thought this might be due to ineffective washing. In this study I tried a better washing technique:
- Incubate polymer membranes, pnc-Si chips, and glass in 1mg/mL BSA for 30min
- Rinse by vigorously swishing in 1500mL PBS for 30s
- Add sample buffer to chips and incubate 30min
- Remove sample buffer and run on a gel
Here is the results of this test:
It looks like pnc-Si adsorbs less protein than the other materials in this study. Note that polymer membranes have larger surface areas (and PES membranes are anisotropic). Also note that the pnc-Si intensities seem similar to the very very first adsorption trial, which was vigorously rinsed in the same manner.
Next step? Repeat studies, perform densitometry, and test alternate samples including oxidized chips, polystyrene, etc.
