w677 Separation

Here are the results of a protein ladder separation with w677.  If you recall, 677 is the 200um wafer that did not fit into the SepCons but had very few pinholes.

S = Standards Ladder, C = Protein solution that never came in contact with membrane, R = Retentate, F = Filtrate

Myosin (Band at top of both retentates) is the only protein really held back by the membrane.  This is similar to previous separations.  The cutoff is higher than 100kD.

The cutoffs are still very high.  In the paper that I hope to do on separations, we need to show two different cutoffs.  This would work for the first cutoff, but it is necessary to find one lower than this too.  Lower would also help the albumin b2microglobulin separation trials that were put on the back burner.

  1. Measured pH in gel box during this experiment to compare with my EO setup (these two setups are very similar-both have Pt wires and buffer solution, but gel is much larger).  Running buffer starts at pH 8.3.  Top measured about 8.5 and bottom measured about 8.0.
