HUVEC on pncSi
In order from left to right:
1) t=0, huvec on pncSi, in EGM (10% FBS)
2) t=24 hrs (cells reach confluence)
3) DIC after staining
4) FITC (live cells)
5) Rhodamine (dead cells)
Its good to see the cells reach confluence and the membranes intact at the same time!
There is a decrease in number of cells in the third image probably because the cells would have fallen off the membrane while rinsing the sample in PBS prior to staining.
Note: live/dead viability kit for mammalian cells was used for staining. It has two components: A- Calcein AM
B- Ethidium homodimer
Its prepared in PBS and takes 30-45 mins in room temperature.
