Discoloration in Sepcon

We have seen that discoloration of chips is slower in Sepcons and this has been speculated to be because of the separation between the ‘well side’ and the ‘membrane side’ of the chip existing in the Sepcon format. To further provide evidence to this theory, I carried out an experiment monitoring the discoloration of samples from W703 (in DMEM w/o serum @ room temp, No RTP) with membrane side up in Sepcons. I used two pnc-Si hybrid transwells and purposely broke the membrane in one of them. In accordance with the theory, the chip with broken membrane discolored much quicker than the intact one as seen in the following:

The membrane is still intact in another sample under same conditions and I am using that sample for bead assay to see how long will it last.
