I repeated the experiment with samples from #703. Two samples were in a dish with one flipped over. Again, the ‘well side up’ sample (flipped) discolored earlier as seen in the 24 hour time point in the figure below.

Having a small, unstirred, confined volume can create all kinds of local effects (pH, Si concentration, temperature, mechanical contact, etc.). Can the sample be raised off the bottom while still facing down?
Yes, I am using sepcon transwell format with membrane side up/down samples in it to check if there is any difference. I was also thinking of using glass beads to raise the chip, but it seems even this format will have trapped environment.
I’d recommend the Sepcon format.
