TEER Testing BBB Device v2.4 with Shrink Wrap and Media
After some changes to my fabrication process, I can report a qualified successful TEER test with media (HBSS) in both channels. I utilized a shrink wrap method of fabrication as seen in a previous post. The test was not perfect because I ruptured the channel while moving the fabricated device to a microscope slide. Also,…
Test Run of SepCon Format
SepCon Experiment (Carrie): Does it work in a SepCon format? Use “Protein A Beads and IgG Binding and Filtering (with SepCon Format)” 9/29/09 Make Buffer B at pH 3 9/30/09 1) Rinse off ethanol 2) Add IgG Take absorbance reading of 1 aliquot of IgG (2×10μl samples) [D7= 0.565abs, D8=0.63abs] Transfer beads to 2ml centrifuge…
Preliminary Test of IgG Binding to Protein A Beads
Last week, Barrett and I made up the three buffers needed to perform this test: Dissociating buffer: 0.1 M glycine-HCl, pH 3.5 1 M Tris Binding buffer: 0.1 M sodium phosphate, 0.15 M NaCl, pH 7.5 In addition, I made a solution of protein A beads that consists of 0.0166 g of beads in 50…
Barcikowski Collaboration Update
Over the last week or so I’ve taken some lovely SEM pictures with Joe, used the light scatter, and ran some separations on filters I assembled myself. Someone from the Barcikowski lab, Dr. Rehbock, got in touch with me and sent me an updated description of the four samples they’re sending and what they want…
High res TEM images @ UR
The image below was taken with UR’s new S/TEM, the FEI Technai F20. It has the capability of resolving lattice images which should benefit the materials development effort. However, we will still rely on Karen’s TEM for most of our imaging since the throughput is higher on that tool.
pinhole-less membranes
Inspired by JP, we decided to try to etch amorphous (unannealed) wafers this week to see if pinhole density changed. To my surprise, the amorphous membranes had no pinholes! Furthermore, I tried to anneal the membranes after the etch and they went flat indicating crystallization! Refer to the table below for the results. Wafer 565:…
