"Phantom" Curves on the TECAN Mess with Reproducibility and Accuracy

    ByKarl Smith March 12, 2013September 24, 2020

At some point, when things settle down, I’ll create a page dedicated to the TECAN machine and document all of the troubleshooting and tricks I’ve learned so far. Before that happens I have another mystery to solve. Take a look at the following three scans:   This first are some scans of BSA filtrates from…

    Micropump Data Update

    ByJascha Wilcox March 12, 2013

After a few more revisions of the micro pump, I performed a significantly more thorough characterization. Flow rate was determined as before using a horizontal capillary of known volume and a stop watch. A extremely small thermocouple was inserted into the PDMS substrate directly adjacent to the fluid outlet channel emptying the…

A quick graph showing that a higher active area/inactive area ratio sees less pore blockage

    ByKarl Smith March 12, 2013

These volume measures were conducted at 5.6 +/-0.2 PSI using the variable area sepcons with either a half slot, one slot, three slots, or the normal five slots. Note that the pressure was normalized via a pressure regulator, and this normalization was confirmed with a manometer. Filtrates were collected in a series of the…

    PEG Microarray Developments

    ByKate Busse March 12, 2013

Updates: We can now put the glass on top of the mask, which allows the use of nonstick agents in the creation of PEG microarrays. The ability to cross-link PEG-Diacrylate (PEG-DA) with a photoinitator and UV exposure was previously tested and shown to work. At the lowest power setting on the Leica (~25%) fluorescence source, we were…

CytoVu Static TEER Device with Yuchen Wang

    ByGreg Madejski March 11, 2013

Yuchen and I have been tasked with integrating TEER electrodes into the Cytovu format. The general idea was to fabricate the TEER electrodes similarly to what I have done already, then add them to the current Cytovu device, using UV ozone bonding and PDMS (to cement in the top electrode). I drew some electrodes to…

40nm Gold Passes Through 40 nm pnc-Si Sepcon Chips; 100nm Fluorospheres Do Not.

    ByKarl Smith March 11, 2013

On Friday Josh and I assembled  Sepcon chips with calculated porosity of ~6.5 %, and avg. pore diameter of ~40 nm into Sepcons.   I was able to pass 40nm gold through the pores (we don’t have any gold larger than that).  Note the “Bad” retentate – for some reason one of the sepcons didn’t filter anything. On…

TECAN absorbance for 96-well plates is topping out even with pure buffer

    ByKarl Smith March 5, 2013

EDIT: This problem was solved thanks to Tejas. It turns out that Falcon BD transparent 96 well plates absorb in the UV range, and are best used for bradford assays or anything that measures absorbance higher than the UV range. Apparently they sell UV-inactive 96 well plates, but they are very expensive. Josh Winans and…
